LOCUS pT11-bs_episome 9985 bp DNA circular 23-JUL-2008 Purpose: can be used for episomal expression in BF of a cassette inserted at the EcoN1 site in the plasmid backbone, but the stability of replication and/or segregation in BF is still less than desirable. pT11bs was made from pT13-11 by substituting the PflMI–EcoN1 fragment of pT13-11 with the corresponding fragment from pHD16-Neo (Hug et al 1993), primarily to provide the BF-compatible Aldolase 3´ UTR. The plasmid was empirically derived by shotgun cloning genomic EcoRI fragments, so the essential region, designated PMS, is a curious chimera of EcoRI fragments from 3 places in the genome. Junction between PMS and Procyclin promoter region was corrected with info from Pradeep on Jun 8 2006. The origin of the plasmid was Hon's pH51, with the PMS being cloned into the RI site, which was lost in cloning.
